QTIP: Quantifier for Topographic Index of Presynaptic-terminal using 3D Confocal Imaging
نویسنده
چکیده
Topography mapping is an important and general strategy for the nervous system to encode the locations of sensory stimuli. It has been difficult to analyze the relative spatial position between adjacent neurons in the same nervous system because of complex fine branches of axon terminals, especially at single-cell resolution. This paper describes an automatic image analysis tool QTIP (Quantifier for Topographic Index of Presynaptic-terminal), which quantifies fine-scale 3D structures’ spatial relative topography. Using confocal microscopic imaging assisted with advanced immunostaining, the presented image quantification tool enables automatic analysis of fine scale topography mapping in a 3D context, exemplified by the axon terminal topography analysis in Drosophila larva somatosensory system. The tool QTIP is developed as an ImageJ plugin and available at http://faculty.cs.niu.edu/~zhou/tool/qtip/.

Imaging of Ca2+ dynamics within the presynaptic terminals of salamander rod photoreceptors.
Although the overall importance of Ca(2+) as a mediator of cell signaling and neurotransmitter release has long been appreciated, the details of Ca(2+) dynamics within the inner segments of vertebrate rod photoreceptors are just beginning to be elucidated. Even less is known regarding Ca(2+) dynamics within the rod presynaptic terminal compartment. Using fura-2 to report changes in intracellula...
متن کاملNile Red Quantifier: a novel and quantitative tool to study lipid accumulation in patient-derived circulating monocytes using confocal microscopy[S]
The inflammatory profile of circulating monocytes is an important biomarker for atherosclerotic plaque vulnerability. Recent research revealed that peripheral lipid uptake by monocytes alters their phenotype toward an inflammatory state and this coincides with an increased lipid droplet (LD) content. Determination of lipid content of circulating monocytes is, however, not very well established....

متن کاملMethods to calibrate and scale axial distances in confocal microscopy as a function of refractive index
Accurate distance measurement in 3D confocal microscopy is important for quantitative analysis, volume visualization and image restoration. However, axial distances can be distorted by both the point spread function (PSF) and by a refractive-index mismatch between the sample and immersion liquid, which are difficult to separate. Additionally, accurate calibration of the axial distances in confo...
